Determination of beta- agonists residues in foodstuff of animal origin by EXTRA and HPLC/MS/MS

Abstract

An Automated Solid Phase Extraction System (EXTRA) and high performance liquid chromatography-tandem mass spectrometry (HPLC/MS/ MS) method was established for the determination of three beta-agonists (clenbuterol, ractopamine, salbutamol) in foodstuff of animal origin. This method uses enzymatic digestion, Samples were extracted using 2 mol/L (pH=5.2) ammonium acetate buffer solution and cleaned up on a all automatic SPE instrument (EXTRA), followed by determination of the residues by HPLC/MS/ MS using atmospheric pressure chemical ionization in the positive ion mode. The recoveries and the RSDs were both meet the requirements. The method was proved to be good reproducibility, high sensitivity, rapid, reliable and suitable for the simultaneous determination of clenbuterol, ractopamine, salbutamol.

Conclusion
The work shown here provides a sensitive and rapid HPLC/MS/MS method with on-line SPE for simultaneous determination of beta-agonists (clenbuterol, ractopamine, salbutamol) in foodstuff of animal origin. The method was proved to be good reproducibility, reliable, the recoveries and the RSDs were both meet the requirements. Therefore, this method is suitable for simultaneous determination of large quantities of samples, and work well for screening the foodstuff of animal origin sold in the market.
